Simple One Step Reaction, Superior Performance
MegaFiTM One-Step RT-PCR is an enzyme mix of OneScript® Hot Reverse Transcriptase and MegaFiTM Fidelity DNA Polymerase with RNaseOFF Ribonuclease Inhibitor, gel loading dye, and all other necessary reagents in a single One-Step 2X RT-PCR Buffer for both highly sensitive and specific reverse transcription and high fidelity PCR amplification in a single reaction tube from any RNA template. It provides flexibility in choosing desired primers for use with a proprietary RT-PCR buffer containing stabilizers and enhancers that optimize the two reactions in a “single step”. This kit offers simple, efficient reaction setup, and is a reliable alternative to conventional “two-step” sequential RT-PCR. Product Features:

MegaFi™ Fidelity DNA Polymerase
MegaFi™ Fidelity DNA Polymerase is the best high fidelity DNA Polymerase on the market. With 1300X better fidelity than regular Taq, this ultrasensitive polymerase amplifies even the most difficult templates.

Ultra-Low Error Rates
MegaFi™ DNA Polymerase is THE BEST high fidelity DNA polymerase. A lacZa-containing sequence was amplified using various DNA polymerases, subcloned, and plated on X-gal-containing media. Colonies without PCR amplified lacZa mutations appear dark blue, while those that have PCR introduced frameshift mutations appear white.
Superior Fidelity

MegaFi™ DNA Polymerase has ~1300X higher fidelity than other enzymes on the market. abm’s MegaFi™ Fidelity DNA Polymerase (Cat. No. G896) has the highest accuracy rate compared to other DNA polymerases as tested using blue/white colony screening. Shown as relative fidelity compared to Taq DNA Polymerase (Taq = 1X). PCR cycle number and mutations resulting in non-phenotypic changes are accounted for.
OneScript® Hot Reverse Transcriptase
Synthesize cDNA from complex RNA templates with abm’s OneScript® Hot Reverse Transcriptase. Engineered for outstanding thermostability, our OneScript® Hot Reverse Transcriptase is ideal for handling secondary structures (e.g. GC-rich templates) as well as long, degraded, or contaminated RNA samples, making it the superior choice for challenging RNA templates.

High cDNA yields from difficult or ultra-low RNA samples
Outstanding Thermostability
OneScript® Hot Reverse Transcriptase has exceptional thermostability. OneScript® Hot Reverse Transcriptase (Cat. No. G593) was used in a 10 min. reaction with RNA template at temperatures ranging from 50 and 80˚C. The resulting synthesized cDNA was followed by PCR and visualized on a 1% agarose gel.
