Recombinant Adeno-Associated Virus (AAV) is the most promising candidate for gene therapy among all viral vectors owing to its non-integrating nature and lack of pathogenicity in animal models. It is also a powerful tool in regenerative medicine due to its broad tropism and ability to transduce both proliferating and quiescent cells.

To qualify for Custom AAV Production without DNA amplification, the following amounts of the AAV vector containing the gene of interest must be supplied at >0.5μg/μl:
- 10^9 GC/ml: 75 μg
- 10^12 GC/ml: 250 μg
- 10^13 GC/ml: 750 μg
For customer-supplied vectors, the customer must indicate prior to the service if any reporter expression will be expected or not. The expected reporter expression, e.g. GFP, should not require induction of expression.

FAQs
- Is your recombinant AAV replication-deficient?
Yes, the replication and capsid genes are provided in trans when the AAV is produced, therefore the packaged virion only has the ITR sequences and the gene of interest. Furthermore, the cis plasmid and rep/cap plasmid do not share any regions of homology, preventing the production of wild-type AAV through recombination system.
- What is the packaging capacity for AAV?
The maximum insert size is < 4.4 kb.
- Is AAV stable? What is the recommended storage temperature?
It is recommended to store AAV at -80˚C for long-term storage. For short-term, AAV is stable at 4˚C for up to three weeks without significant loss of activity.
- How much AAV virus should be submitted for the Custom AAV Titration service, Cat# C238?
Please send at least 50μl of virus if it is low titer. If it is expected to be high titer AAV virus, please send at least 20μl of sample.
