Tips: recommend to optimize the reactions for each primer set and determining the ideal time for the reaction to be ran for.

1. Determine optimal temperature. Using a temperature gradient for a set time, evaluate the quality of bands and if they follow the expected. Temperatures can range from 55°C to 72°C for reaction runs.
2. Determine the optimal reaction time. High or low copy number of amount of template input will influence the time it takes to see a positive reaction. Recommend to start with 15 minute intervals. Check on your NTC as with longer reaction times the control is susceptible to false positive. Confirm results of amplification of the NTC by running product on agarose gel and evaluate the banding pattern.
