Description T5 exonuclease catalyzes the degradation of nucleotides either from the 5’ termini or at nicks of linear or circular dsDNA in a 5’ to 3’ direction. This exonuclease also exhibits ssDNA endonuclease activity in the presence of magnesium ions, but will not degrade supercoiled dsDNA.

Applications
  • Degradation of ssDNA, dsDNA linear, circular or nicked, while preserving supercoiled plasmid DNA.
  • Removal of contaminant denatured DNA isolated during alkaline lysis plasmid purification.
  • Generation of 3’ overhangs for cloning procedures.
Concentration 10000 U/ml
Format Enzyme supplied with a 10X Reaction Buffer. Incubate at 37°C.
Storage Condition Store all components at -20°C.
Storage Buffer 50 mM Tris-HCl (pH 7.5), 100 mM NaCl, 1 mM DTT, 0.1 mM EDTA, 50% Glycerol and 0.1% Triton X-100.
Note One unit is defined as the amount of enzyme necessary to catalyze the release of 1 nmol of acid-soluble nucleotides from double-stranded DNA in a 50 µl volume in 30 minutes at 37°C. This product is distributed for laboratory research only. Caution: Not for diagnostic use.
